Heteromultimeric Assembly of Human Potassium Channels

چکیده
To gain insight into the molecular basis of cardiac repolarization, we have expressed K' channels cloned from ventricular myocardium in Xenopus oocytes. A recently identified human cardiac K' channel isoform (human Kvl.4) has properties similar to the 4-aminopyridine-sensitive calcium-insensitive component of the cardiac transient outward current. However, these channels recovered from inactivation much slower than native channels. Hybrid channels consisting of subunits from different K' channel clones (delayed rectifier channels [Kvl.l, Kv1.2, and Kvl.5] and Kvl.4) were created by coinjection of cRNAs in oocytes. Multimeric channels consisting of Kvl.4:Kvl.1, Kvl.4:Kvl.2, and Kvl.4:Kvl.5 were expressed and compared. The hybrid channels displayed characteristics of heterotetrameric channels with kinetics that

Dynamic subunit stoichiometry confers a progressive continuum of pharmacological sensitivity by KCNQ potassium channels.
Voltage-gated KCNQ1 (Kv7.1) potassium channels are expressed abundantly in heart but they are also found in multiple other tissues. Differential coassembly with single transmembrane KCNE beta subunits in different cell types gives rise to a variety of biophysical properties, hence endowing distinct physiological roles for KCNQ1-KCNEx complexes. Mutations in either KCNQ1 or KCNE1 genes result in...
متن کاملCardiac small conductance Ca2+-activated K+ channel subunits form heteromultimers via the coiled-coil domains in the C termini of the channels.
RATIONALE Ca(2+)-activated K(+) channels are present in a wide variety of cells. We have previously reported the presence of small conductance Ca(2+)-activated K(+) (SK or K(Ca)) channels in human and mouse cardiac myocytes that contribute functionally toward the shape and duration of cardiac action potentials. Three isoforms of SK channel subunits (SK1, SK2, and SK3) are found to be expressed....
متن کاملSubunit assembly and domain analysis of electrically silent K+ channel alpha-subunits of the rat Kv9 subfamily.
Alpha-subunits of the voltage-gated potassium channel (Kv) subfamily Kv9 show no channel activity after homomultimeric expression in heterologous expression systems. This report shows that heteromultimeric expression of rKv9.1 and rKv9.3 specifically suppresses the currents mediated by alpha-subunits of the Kv2 and Kv3 subfamilies but does not affect currents mediated by alpha-subunits of the K...
متن کاملHeteromultimeric potassium channels formed by members of the Kv2 subfamily.
Four alpha-subunits are thought to coassemble and form a voltage-dependent potassium (Kv) channel. Kv alpha-subunits belong to one of four major subfamilies (Kv1, Kv2, Kv3, Kv4). Within a subfamily up to eight different genetic isotypes exist (e.g., Kv1.1, Kv1.2). Different isotypes within the Kv1 or Kv3 subfamily coassemble. It is not known, however, whether the only two members of the vertebr...
متن کاملA mechanism for combinatorial regulation of electrical activity: Potassium channel subunits capable of functioning as Src homology 3-dependent adaptors.
It is an open question how ion channel subunits that lack protein-protein binding motifs become targeted and covalently modified by cellular signaling enzymes. Here, we show that Src-family protein tyrosine kinases (PTKs) bind to heteromultimeric Shaker-family voltage-gated potassium (Kv) channels by interactions between the Src homology 3 (SH3) domain and the proline-rich SH3 domain ligand seq...
